Though I am looking forward for some mythology reveals in the next 4 episodes, I am truly excited for the present day questions. Not only are they the ones most likely to answer (who's going to get more screen time, Locke or Walt?), but they're the ones with the most set up and likely will be the ones with the greatest answers. Lets go, folks!
1. Jacob's Replacement
Jack - Most likely, he's had the largest character development from Man of Science to Man of Faith.
Hurley - Personally, I think he'd be a great replacement of Richard, if the spot's up for grabs.
Sawyer - Poor odds. I can't imagine him living in a stone foot for eternity, and even though Sawyer has proven he can take leadership, he doesn't care about the island.
Sun/Jin - Do you think they can work as a team? If one of them were to snuff it I could see it happening, but otherwise I don't think they'd leave their families from home. But what if Kwon referred to Ji-Yeon?
Sayid - Doubtful, as he is part evil.
Locke - He's alive! Joking aside, Locke would be perfect for the part if he were to somehow come back via FST/OT-crossover-ressurection-type-thing.
Ben - The underdog, it'd be interesting if Jacob gave him a second chance. The main reason I like this possibility is because it'd round out Ben's storyline; while his performance in Dr. Linus was moving, that'd be a sad way to end the story of devious, manipulative Ben.
Kate - Crossed out in the Cave, but not the Lighthouse? Any significance, seeing as MIB has apparently only visited the former?
Desmond - Is this time-traveller an unknown candidate? His role in the FST seems to mirror Jacob in the flashbacks.
Frank - If Illana knew the 6 candidates, why would she insist on taking Frank along for the ride at the end of Season 5?
Here's my list, from most likely to least likely:
2. Vanquisher of the Man in Black
Jack - Though he seems to be the best positioned as of now, does he have the stuff?
Sayid - Could MIB be losing his grip on Sayid? Especially after the questionable murder of Desmond.
Claire - Same reasons as Sayid above. What's more important to her in her crazy state of mind, MIB or Aaron?
Richard - He obviously would if given the chance, but wouldn't MIB make minced meat out of him?
Ben - Wouldn't it be ironic if Ben murdered MIB with a great dagger on Jacob's behalf? Or just plain silly?
Locke - He's alive! I seem to be stuck on the resurrection of OT Locke, as unlikely as it might be. I just think it'd be great to prove that when FST/OT crossovers are involved, dead is not dead.
Desmond - Is his agenda only to bridge the two timelines, or does he know the significance of the island conflict?
Widmore - A minor character with some serious firepower.
Miles - I read a theory that reminded me that Miles is the only one not to have spoken to MIB. If it is true that 'once you have spoken to him, it's over', it just may be that Miles serves a purpose in this season besides comic relief.
Zoe - Lol, just for kicks.
Himself - Accidental or on purpose?
Jacob - Will Jacob make a return? Let's hope so/not.
Nobody - Will evil triumph over good? Or will MIB see a fate worse than death?
Other - Though Lost may have many a main character, I cannot imagine any of the other main cast plunging a dagger into MIB's chest; none of them are too deep in the Lost mythology.
And the verdict:
3. The Other Outrigger (You remember, right? After Sawyer et. all flash to the beach camp with post-Ajira Airway flight, they have an ocean shoot-out with an unknown group.)
Richard, Ben, and Miles - We know they plan on stocking up at the Barracks, and then heading for Hydra Island. We also know Ben used one of the remaining outriggers just a week earlier. (If my mind's timeline is correct)
Widmore redshirts - It'd be an easy out if the writer's were stuck in a corner. Plus, maybe Zoe would kick the can!
Losties on Hydra Island - This include Kate, Sun, Jin, Sawyer, Claire, Frank, and Hurley. Perhaps they travel back to the island in an escape from Widmore? Mirroring Season 3?
MIB's Faction - Includes MIB, Jack, Sayid, and background Others. Perhaps travelling to Hydra Island? Maybe Cindy Chandler gets what's coming to her?
Other - Desmond? Boone? Dogen? Resurrected Locke? Who knows?
Unsolved - Will the writers forget it? I doubt it, considering that it was a scene that wasn't exactly necessary from last year, when they were probably structuring the final stages of the show.

4. Who Will Die? No explanations, just my suggested list from most to least likely. No one is safe. Doesn't include the Flash-Sideways Timeline.
