Why, you might ask, do you say Desmond is the main character of LOST?
(Hint: It's not his Scottish accent, though that helps)

Catalyst of Main Events

First of all, Desmond causes all the big events of the show
- He leaves on Widmore's race around the world, and Penny starts looking for him (this will be important in the last bullet point)
- He caused the crash of Oceanic Flight 815 ("Live Together, Die Alone, Part 1")
- He saved the world a bunch of times, in the past (throwing away the ring ("Live Together, Die Alone, Part 1")) and the present (pushing the button, even after Kelvin's death)
- He renewed John Locke's determination to open 'the hatch' ("Deus Ex Machina")
- He turned the Failsafe Key and saved the world ("Live Together, Die Alone, Part 1")
- He predicted and preempted Charlie's death multiple times, as well as Naomi's arrival- and because they found Naomi before Mikhail, she lived long enough to call her freighter- and season 4 happened
- He kept the boat un-exploded enough to rescue the O6, and himself, and to have an awesome happy ending with Penny ("There's No Place Like Home, Part 2")
- Because Penny was looking for him, she rescued the O6 ("There's No Place Like Home, Part 2")
Connection to Main Bad Guy

Many main characters have some sort of connection to the main bad guy.
Mr. Widmore was introduced to us through Desmond in season 2. Obviously their link will become more important in the following (and final) seasons

- Desmond is awesome. It doesn't mean he's the main character, but it helps

Through the above evidence, and more, I have come to believe that the show LOST revolves around Desmond as the main character. It may be seem to be Jack or a host of others and Others and freighties, but it's Des that takes the cake. His story of redemption that covers the arc of the entire show, and his compelling and very real love story with Penny... his distinct separation from being either a crash survivor or an Other or some other faction... Desmond is his own faction.
The story arc seems to follow him... from the mysteries of DHARMA to the crash of the plane to the Failsafe Key to his future-flashes determing the fate of the rest of the show to his interactions with the electromagnetic physics of the Island to his fateful link to Charles Widmore... I do think Desmond is the main character of LOST.
