  • The "drug" is not a drug at all, but a concoction of dirt and foliage from the Island. In Boone's case the substance is applied to an open wound, and in Locke's case eaten. The user is ingesting the "soul" of the Island.
  • During John's encounter with the Smoke Monster, MIB gave him the know-how on the potion. We know that the Smoke Monster gives flashes to certain people. I believe and I don't think I'm alone in saying, that the Smoke Monster gave John a "reflection" on the whole Island. John sure looked pretty sure of himself when he was mixing that potent potion up. Almost as if he knew exactly what he was doing, like he was shown how!
